A method of efficiently constructing a reproducible animal by using a totipotent cell having a mitochondrial DNA (for example, wilt type DNA) compatible with the nuclear DNA into its mitochondrial DNA via transfer or substitution; and an animal constructed thereby. In the case where the totipotent cell is an ES cell originating in an inbred strain mouse, it is desirable to employ the tetraploid rescue method. In constructing a chimeric animal, the mitochondrial DNA originating in a totipotent cell of the animal employed is substituted by a wild type mitochondrial DNA by the backcross mating method, the nuclear substitution method or the like and then injected into a tetraploid fertilized egg. Thus, a reproducible inbred strain chimeric animal is constructed while avoiding the risk of the death of the obtained animal due to respiratory disturbance, etc. immediately after birth. The reproducible inbred strain chimeric animal thus obtained is usable in gene function analyses, animal experiments, etc. without resort to troublesome inbred strain procedures.
L'invention concerne un procédé de construction efficace d'un animal reproductif au moyen d'une cellule totipotente contenant un ADN mitochondrial (par exemple un ADN de type sauvage) compatible avec l'ADN nucléaire dans son ADN mitochondrial par transfert ou substitution, ainsi que l'animal construit par ce procédé. Dans le cas où la cellule totipotente est une cellule souche embryonnaire provenant d'une souris de souche pure, il est souhaitable d'utiliser la méthode de sauvetage par tétraploïdie. Dans la construction d'un animal chimère, l'ADN mitochondrial d'une cellule totipotente de l'animal utilisé est substitué par un ADN mitochondrial de type sauvage au moyen du procédé de rétrocroisement, du procédé de substitution nucléaire ou analogue, puis injecté dans un oeuf tétraploïde fertilisé. Ainsi, un animal chimère reproductif de souche pure est construit tout en évitant le risque de la mort de cet animal due à un trouble respiratoire, ou autre, immédiatement après sa naissance. L'animal chimère reproductif de souche pure ainsi obtenu peut être utilisé dans des analyses de fonctions génétiques, des expérimentations animales, et d'autres essais, sans recourir à des procédures de souche pure problématiques.
