Methods and compositions are provided for the creation and screening of non- human animal models having many of the histologic characteristics of human psoriasis. Immunocompromised host animals are injected with a purified population of CD45Rb positive cells, which are tolerant of the host major histocompatibility antigens, but are mismatched at one or more minor antigens. The injected cells are stimulated with a pro-inflammatory cytokine, e.g. IL-12, and a polyclonal activating agent. The injected animals develop a chronic skin disorder that includes histological features observed in human psoriasis, e.g. rete pegs, severe acanthosis and infiltration of Th1 cells into the dermis.
L'invention se rapporte à des méthodes et à des compositions permettant la création et le tri de modèles animaux, non humains, présentant de nombreuses caractéristiques histologiques du psoriasis humain. On injecte dans des animaux hôtes immunodéprimés une population purifiée de cellules positives CD45Rb, qui tolèrent les antigènes majeurs d'histocompatibilité des hôtes, mais sont non-appariés à au moins un des antigènes mineurs. Les cellules injectées sont stimulées par une cytokine pro-inflammatoire, par exemple IL-12, et un agent d'activation polyclonal. Les animaux injectés développent un trouble cutané chronique qui présente certaines caractéristiques histologiques du psoriasis humain, par exemple, des pédicelles d'insertion, une hyperacanthose et une infiltration des cellules Th1 dans le derme.
