The present invention relates to a non-human animal model, cell and tissue cultures derived therefrom, which do not produce or produce only suboptimal levels of one or more functional sestrins and in addition do not produce or do only produce suboptimal levels of latent transforming growth factor ß binding protein 4 (ltbp4). Furthermore, the present invention relates to a method for selecting agents for treating the pulmonary emphysema and/or the colorectal cancer exhibited by utilizing the animal model, cell or tissue culture of the invention. The animal model, cell or tissue culture is suitable for preclinical testing of efficacy, toxicity and bioavailability of potential agents.
L'invention porte sur un modèle animal non humain, et sur des cultures cellulaires et tissulaires dérivées de ce dernier, qui ne produisent pas ou ne produisent que des niveaux sous-optimaux d'une ou plusieurs sestrines fonctionnelles et qui, en outre, ne produisent pas ou ne produisent que des niveaux sous-optimaux de la protéine de liaison 4 du facteur de croissance de transformation latent ß ("latent transforming growth factor ß binding protein 4" ou ltbp4). Par ailleurs, l'invention concerne un procédé qui permet de sélectionner des agents destinés au traitement de l'emphysème pulmonaire et/ou du cancer colorectal au moyen du modèle animal et de la culture cellulaire ou tissulaire de l'invention. Le modèle animal et la culture cellulaire ou tissulaire précités sont aptes à permettre une vérification préclinique de l'efficacité, de la toxicité et de la biodisponibilité d'agents potentiels.
