The present invention relates to an annealing control primer for improving annealing specificity in nucleic acid amplification and its applications to all fields of nucleic acid amplification-involved technology. The present primer comprises (a) a 3'-end portion having a hybridizing nucleotide sequence substantially complementary to a site on a template nucleic acid to hybridize therewith; (b) a 5'-end portion having a pre-selected arbitrary nucleotide sequence; and (c) a regulator portion positioned between said 3'-end portion and said 5'-end portion comprising at least one universal base or non- discriminatory base analog, whereby said regulator portion is capable of regulating an annealing portion of said primer in association with annealing temperature.
L'invention concerne une amorce de commande de renaturation servant à améliorer la spécificité de renaturation dans l'amplification d'acides nucléiques, ainsi que ses applications à tous les domaines de technologie impliquant l'amplification d'acides nucléiques. L'amorce selon l'invention comprend : (a) une partie d'extrémité 3' présentant une séquence nucléotidique d'hybridation pratiquement complémentaire d'un site sur un acide nucléique matrice pour l'hybridation ; (b) une partie d'extrémité 5' présentant une séquence nucléotidique arbitraire présélectionnée et (c) une partie régulatrice située entre la partie d'extrémité 3' et la partie d'extrémité 5' et comprenant au moins un analogue de base universelle ou de base non discriminatoire. Selon l'invention, cette partie régulatrice est apte à réguler une partie de renaturation de l'amorce, en association avec la température de renaturation.
