The invention provides a Vitaxin antibody and a LM609 grafted antibody exhibiting selective binding affinity to .alpha.v.beta.3. The Vitaxin antibody consists of at least one Vitaxin heavy chain polypeptide and at least one Vitaxin light chain polypeptide or functional fragments thereof. Also provided are the Vitaxin heavy and light chain polypeptides and functional fragments. The LM609 grafted antibody consists of at least one CDR grafted heavy chain polypeptide and at least one CDR grafted light chain polypeptide or functional fragment thereof. The invention additionally provides a high affinity LM609 grafted antibody comprising one or more CDRs having at least one amino acid substitution, where the .alpha.v.beta.3 binding activity of the high affinity LM609 grafted antibody is enhanced. Nucleic acids encoding Vitaxin and LM609 grafted heavy and light chains as well as nucleic acids encoding the parental non-human antibody LM609 are additionally provided. Functional fragments of such encoding nucleic acids are similarly provided. The invention also provides a method of inhibiting a function of .alpha.v.beta.3. The method consists of contacting .alpha.v.beta.3 with Vitaxin or a LM609 grafted antibody or functional fragments thereof under conditions which allow binding to .alpha.v.beta.3. Finally, the invention provides for a method of treating an .alpha.v.beta.3-mediated disease. The method consists of administering an effective amount of Vitaxin or a LM609 grafted antibody or functional fragment thereof under conditions which allow binding to .alpha.v.beta.3.
L'invention concerne un anticorps vitaxine et un anticorps greffé LM609 présentant une affinité de liaison sélective avec .alpha.¿v?.beta.¿3?. L'anticorps vitaxine comporte au moins un polypeptide vitaxine à chaînes lourdes et au moins un polypeptide vitaxine à chaînes légères ou des fragments fonctionnels desdits polypeptides. L'invention concerne également des polypeptides vitaxine à chaînes lourdes et légères et des fragments fonctionnels. L'anticorps greffé LM609 comprend au moins un polypeptide à chaînes lourdes greffé sur la région déterminante de la complémantarité (CDR) et au moins un polypeptide à chaînes légères greffé sur la région CDR ou des fragments fonctionnels desdits polypeptides. L'invention a en outre pour objet un anticorps greffé LM609 à haute affinité comprenant une ou plusieurs régions CDR présentant au moins une substitution d'acide aminé, ce qui favorise l'activité de liaison .alpha.¿v?.beta.¿3? de l'anticorps greffé LM609 à haute affinité. L'invention porte encore sur des acides nucléiques codant les chaînes lourdes et légères greffées vitaxine et LM609 et des acides nucléiques codant l'anticorps LM609 non humain parent, de même que des fragments fonctionnels de tels acides nucléiques codants. L'invention concerne aussi un procédé permettant d'inhiber une fonction de .alpha.¿v?.beta.¿3?. Ce procédé consiste à mettre .alpha.¿v?.beta.¿3? en contact avec un anticorps greffé LM609 ou vitaxine ou avec des fragments fonctionnels de ces anticorps dans des conditions permettant la liaison avec .alpha.¿v?.beta.¿3?. Enfin, l'invention concerne un procédé de traitement de maladies induites par .alpha.¿v?.beta.¿3?. Ce procédé consiste à administrer une dose efficace d'un anticorps greffé LM609 ou vitaxine ou des fragments de ces anticorps dans des conditions permettant la liaison avec .alpha.¿v?.beta.¿3?.
