The present invention provides a recombinant immunogenic obtained by tandem multimerization of a B-cell epitope bearing fragment of A.beta.42, within the active loop site of a carrier (display site), preferably bacterial thioredoxin (Trx). Polypeptides bearing multiple copies of A.beta.42 fragments, preferably with an interposed amino acid linker, were constructed and injected into mice in combination with an adjuvant. Elicited antibodies were found to selectively bind to fibrillar and/or oligomers A.beta. within neuritic AD plaques.

La présente invention concerne un immunogène recombinant obtenu par multimérisation en tandem d'un fragment d'A.beta.42 portant un épitope de lymphocyte B dans le site de boucle active d'un porteur (site de présentation), de préférence la thiorédoxine (Trx) bactérienne. Des polypeptides portant de multiples copies de fragments d'A.beta.42, de préférence avec un segment d'acides aminés de liaison intercalé, ont été assemblés et injectés à des souris en combinaison avec un adjuvant. Les anticorps produits se lient sélectivement à l'A.beta. fibrillaire et/ou oligomère à l'intérieur des plaques séniles de la maladie d'Alzheimer.
