This invention relates to new anti-EGFR antibodies and single-chain Fvs (scFvs) thereof which can be obtained from phage-antibody libraries constructed from cells of an immunized mammalian, preferably a mouse. Two of the single-chain Fvs isolated from the phage- antibody libraries were engineered to create partially humanized whole antibody molecules These chimeric anti-EGFR antibodies contain constant regions of human immunoglobulins and can be used as well as the single-chain Fvs as agents for the diagnosis and therapy of human tumors.

L'invention se rapporte à des nouveaux anticorps anti-récepteurs du facteur de croissance épidermique (EGFR) ainsi qu'à des fragments (Fvs) à simple chaîne (scFvs) de ceux-ci que l'on peut obtenir à partir de bibliothèques d'anticorps-phages assemblés à partir de cellules d'un mammifère immunisé, de préférence une souris. On a manipulé génétiquement deux des Fvs à simple chaîne isolés de bibliothèques d'anticorps-phages afin de créer des molécules d'anticorps complet, partiellement humanisées. Ces anticorps chimères anti-EGFR contiennent des régions constantes d'immunoglobulines humaines, et peuvent être utilisés en même temps que les Fvs à simple chaîne en tant qu'agents de diagnostic et de thérapie de tumeurs humaines.
