The present invention discloses anti-idiotypic antibodies and fragments thereof against inhibitory Factor VIII antibodies, said inhibitory antibodies having an affinity for the C2 domain of Factor VIII. The anti-idiotypic antibodies of the present invention are able to completely neutralise in vitro and in an in vivo mouse model the inhibitory activity of FVIII inhibitors. The anti idiotypic antibodies of the present invention can he applied for the prevention, treatment or reduction of bleeding disorders of hemophilia patients with inhibitory antibody against the C2 domain of Factor VIII.
La présente invention concerne des anticorps anti-idiotypiques et des fragments de ceux-ci permettant de lutter contre des anticorps inhibiteurs du facteur VIII. Ces anticorps inhibiteurs présentent une affinité pour le domaine C2 du facteur VIII. Les anticorps anti-idiotypiques selon cette invention peuvent neutraliser complètement in vitro et chez un modèle de souris in vivo l'activité d'inhibition des inhibiteurs du FVIII et peuvent être utilisés pour prévenir, traiter ou atténuer des troubles de saignement chez des patients hémophiles, à l'aide d'un anticorps inhibiteur permettant de lutter contre le domaine C2 du facteur VIII.
