Antimicrobial agents that can serve as replacements to conventional pharmaceutical antibiotics are disclosed. The antimicrobial agents comprise conjugatively transmissible plasmids that kill targeted pathogenic bacteria, but are not harmful to donor bacteria. Two types of lethal transmissible plasmids are disclosed. One type kills recipient bacteria by unchecked ("runaway") replication in the recipient cells and is prevented from occuring in donor cells. Another type kills recipient bacteria by expressing a gene that produces a product detrimental or lethal to recipient bacterial cells, that gene being prevented from expression in donor cells.
L'invention concerne des agents antimicrobiens susceptibles de remplacer des antibiotiques pharmaceutiques classiques. Ces agents antimicrobiens comprennent des plasmides transmissibles de manière conjuguée qui tuent les bactéries pathogènes cibles sans nuire aux bactéries donatrices. L'invention concerne deux types de plasmides transmissibles létaux. L'un tue les bactéries receveuses par réplication non contrôlée (autoamplifiable) dans les cellules receveuses, tout en étant empêché de survenir dans des cellules donatrices. L'autre tue les bactéries receveuses en exprimant un gène qui fabrique un produit nuisible ou létal pour les cellules bactériennes receveuses, tout en empêchant ledit gène de s'exprimer dans des cellules donatrices.
