This invention provides novel methods of obtaining autologous monoclonal antibodies (AMABs) to self-antigens or homologs thereof. The method involves obtaining a genetically engineered host animal that does not biosynthesize at least one epitope of the antigen and utilizes the lack of self-tolerance of the host to the epitope to produce antibodies specific to the antigen. The invention also encompasses the AMABs produced by the methods. The invention further encompasses methods of isolating cells comprising the use of such AMABs that have specificity for a cell surface antigen.

Cette invention fournit de nouvelles méthodes destinées à la production d'anticorps monoclonaux autologues (AMA) dirigés contre des antigènes autologues ou leurs homologues. La méthode comprend l'obtention par génie génétique d'un animal hôte qui ne biosynthétise pas au moins un épitope de l'antigène et qui tire profit de son absence de tolérance envers l'épitope pour produire des anticorps spécifiques à l'antigène. L'invention englobe également les AMA produits grâce aux dites méthodes ainsi que des méthodes d'isolement de cellules, notamment l'utilisation de ceux parmi les AMA qui présentent une spécificité pour un antigène à la surface des cellules.
