An antibody binding to IGF-IR and inhibiting the binding of IGF-I and IGF-II to IGF-IR which is characterized in that said antibody is a) is ofIgG1 isotype, b) shows a ratio of IC50 values of inhibition of the binding of IGF-I to IGF-IR to the inhibition of binding of IG-II to IGF-IR of 1:3 to 3:1, c) inhibits for at least 80% at a concentration of 5 nM IGF-IR phospohrylation in a cellular phosphorylation assay using 3T3 cells providing 400,000 to 600,000 molecules IGF-IR per cell in a medium containing 0.5% heat inactivated fetal calf serum (FCS) when compared to such an assay without said antibody, and d) shows no IGF-IR stimulating activity measured as IGF-IR phophorylation at a concentration of 10 ~M in a cellular phosphorylation assay using 3T3 providing 400,000 to 600,000 molecules IGF-IR per cell in a medium containing 0,5% heat inactivated fetal calf serum (FCS) when compared to such an assay without said antibody has improved properties in antitumor therapy.

L'invention concerne un anticorps se liant au récepteur I du facteur de croissance de type insuline (IGF-IR) et inhibant la liaison du IGF-I et du IGF-II au récepteur IGF-IR. L'invention est caractérisée en ce que cet anticorps est a) de l'isotype IgG1, b) présente un rapport entre les valeurs IC¿50 ?d'inhibition de la liaison de IGF-I au IGF-IR et l'inhibition de la liaison du IGF-II au IGF-IR de 1:3 à 3:1, c) inhibe pour au moins 80 % à une concentration de 5 nM IGF-IR, la phosphorylation dans une analyse de phosphorylation cellulaire utilisant des cellules 3T3 fournissant 400,000 à 600,000 molécules de IGF-IR par cellule dans un milieu contenant 0.5 % de sérum embryonnaire de veau (FCS) inactivé par la chaleur comparé à une telle analyse sans cet anticorps, et d) ne présente aucune activité de stimulation du IGF-IR mesurée en tant que phosphorylation du IGF-IR à une concentration de 10 ?M dans une analyse de phosphorylation cellulaire utilisant des cellules 3T3 fournissant 400,000 à 600,000 molécules de IGF-IR par cellule dans un milieu contenant 0,5 % de sérum embryonnaire de veau (FCS) inactivé par la chaleur comparé à une telle analyse sans cet anticorps. Cet anticorps a des propriétés améliorées dans le traitement antitumoral.
