An antibody binding to human IL-1R and inhibiting the binding of human IL-1 to IL-1R, characterized in that said antibody is obtainable from hybridoma cell line MAK<h-IL-1RI>2D8 (DSM ACC 2601) or is a chimeric, humanized or T cell epitope depleted antibody variant or a fragment of said antibody showing an IC50 value of 35 pM or lower for the inhibition of IL-1 mediated secretion of Il-8 and IL-6 in human fibroblast cells like MRC5 (ATCC CCL 171) has advantageous properties for the treatment of inflammatory diseases.
L'invention concerne un anticorps se liant au IL-1R humain et inhibant la liaison du IL-1 humain au IL-1R. L'invention est caractérisée en ce que cet anticorps peut être obtenu d'une lignée cellulaire d'hybridome MAK<h-IL-1RI>2D8 (DSM ACC 2601), ou bien c'est une variante d'anticorps appauvri chimérique, humanisé ou épitope de cellule T, ou bien encore un fragment de cet anticorps d'une valeur IC50 inférieure ou égale à 35 pM pour inhiber la sécrétion associée au IL-1 de Il-8 et de IL-6 dans des cellules fibroblastes humaines telles que MRC5 (ATCC CCL 171). L'anticorps de l'invention présente des propriétés intéressantes pour le traitement de maladies inflammatoires.
