The invention includes antibodies that provide superior anti-coagulant activity by binding native human TF with high affinity and specificity. Antibodies of the invention can effectively inhibit blood coagulation in vivo. Antibodies of the invention can bind native human TF, either alone or present in a TF: VIIa complex, effectively preventing factor X binding to TF or that complex, and thereby reducing blood coagulation. Preferred antibodies of the invention specifically bind a conformational epitope predominant to native human TF, which epitope provides an unexpectedly strong antibody binding site.
La présente invention concerne des anticorps qui déploient une activité anticoagulante supérieure en se liant à un facteur III natif humain avec une grande affinité et une grande spécificité. Les anticorps de la présente invention peuvent inhiber avec efficacité la coagulation sanguine in vivo. Les anticorps de la présente invention peuvent se lier à un facteur III natif humain seul ou présent dans un complexe facteur III: VIIa, empêchant effectivement le facteur X de se lier au facteur III ou au complexe, et réduisant de la sorte la coagulation sanguine. Les anticorps préférés de la présente invention se lient spécifiquement à un épitope de conformation prédominant dans le facteur III natif humain, épitope qui constitue un site de fixation d'anticorps inopinément fort.
