The invention provides an immunoglobulin molecule which binds SI(KDR) with an affinity comparable to human VEGF, and that neutralizes activation of KDR. Immunoglobulin molecules include monovalent single chain antibodies, multivalent single chain antibodies, diabodies, triabodies, antibodies, humanized antibodies and chimerized antibodies. The invention further provides nucleic acid molecules that encode these immunoglobulin molecules. The invention also provides a method of making the immunoglobulin molecules mentioned above. The invention further provides a method of neutralizing the activation of KDR, a method of inhibiting angiogenesis in a mammal and a method of inhibiting tumor growth in a mammal with such immunoglobulin molecules.
La présente invention concerne une molécule d'immunoglobuline se liant au KDR avec une affinité comparable au VEGF humain, et neutralisant l'activation de KDR. Les molécules d'immunoglobulines contiennent des anticorps à chaîne unique monovalents, des anticorps à chaîne unique multivalents, des dianticorps, trianticorps, anticorps, et des anticorps humanisés et chimérisés. Par ailleurs, cette invention concerne des molécules d'acide nucléique codant ces molécules d'immunoglobulines. De plus, cette invention concerne un procédé de préparation des molécules d'immunoglobulines précitées. L'invention concerne également un procédé de neutralisation de l'activation de KDR, un procédé d'inhibition de l'angiogenèse chez un mammifère ainsi qu'un procédé d'inhibition de croissance tumorale chez un mammifère utilisant ces molécules d'immunoglobulines.
