Antibodies specifically recognizing a distinct population of human dendritic cells (DCs) and methods of isolating said DCs using said antibodies. Furthermore, antigens and epitopes recognized by the above-described antibodies as well as polynucleotides encoding said antibodies. Also vectors comprising said polynucleotides as well as host cells transformed therewith and their use in the production of said antibodies. Additionally, polypeptides comprising a domain of the binding site of the aforementioned antibodies, or an antigen or epitope described above and at least one further, preferably functional domain as well as polynucleotides encoding such polypeptides. Furthermore, vectors comprising said polynucleotides, host cells transfected with said polynucleotide or vector and their use for the preparation of the above-described polypeptides. Further a method for isolating or identifying DCs as defined above as well as DCs obtainable by said method and/or characterized by recognition of the above-described antibody, and/or containing the aforementioned antigen or epitope. Moreover, a method for preparing or identifying T cells in a certain status as well as methods for identifying compounds which interfere with T cell mediated activation of immune responses. In addition kits, and compositions, preferably pharmaceutical and diagnostic compositions are provided comprising any of the aforedescribed antibodies, antigens, epitopes, polypeptides, polynucleotides, vectors, dendritic cells or T cells or compounds obtainable by the aforementioned method.
L'invention concerne des anticorps qui reconnaissent spécifiquement une population distincte de cellules dendritiques (DC) humaines et des procédés permettant d'isoler ces cellules dendritiques à l'aide desdits anticorps. L'invention concerne également des antigènes et des épitopes reconnus par ces mêmes anticorps, ainsi que des polynucléotides codant pour ces anticorps. L'invention concerne en outre des vecteurs comprenant ces polynucléotides ainsi que des cellules hôtes transformées avec des derniers et l'utilisation de ces cellules hôtes pour la production des anticorps décrits. Sont également décrits, des polypeptides comprenant un domaine du site de liaison desdits anticorps, ou un antigène ou un épitope décrit ci-dessus, et au moins un autre domaine, de préférence fonctionnel, ainsi que des polynucléotides codant pour de tels polypeptides. L'invention concerne encore des vecteurs comprenant ces polynucléotides, des cellules hôtes transfectées avec ces polynucléotides ou vecteurs, et leur utilisation dans la préparation des polypeptides décrits ci-dessus. Relèvent également de l'invention un procédé permettant l'isolement ou l'identification des cellules dendritiques décrites ci-dessus et les cellules dendritiques obtenues par ce procédé et/ou caractérisées par une reconnaissance par l'anticorps décrit, et/ou contenant l'antigène ou l'épitope décrits. A cela s'ajoutent un procédé de préparation ou d'identification de lymphocytes T dans un certain état, ainsi que des procédés d'identification de composés interférant avec l'activation par les lymphocytes T des réponses immunitaires, ainsi que des kits et des compositions, de préférences des compositions pharmaceutiques et diagnostiques, comprenant n'importe lesquels des anticorps, antigènes, épitopes, polypeptides, polynucléotides, vecteurs, cellules dendritiques ou lymphocytes T décrits, ou des composés obtenus par le procédé décrit précédemment.
