Disclosed are antibody fusion proteins with a modified FcRn binding site and nucleic acid molecules encoding them. The antibody fusion protein include two polypeptide chains, wherein the first polypeptide chain includes a biologically, preferably therapeutically active molecule linked to at least a portion of an immunoglobulin constant region. The second polypeptide chain includes at least a portion of an immunoglobulin constant region. One of the polypeptide chains includes a mutation in the constant region that results in reduction or loss of FcRn binding but does not cause substantial reduction of serum half-life of said antibody fusion protein. In a preferred embodiment the invention relates to respectively constructed Fc-IFN.beta. molecules. Also disclosed are methods of producing the fusion proteins and methods of using the fusion proteins for treating diseases and conditions alleviated by the administration of the fusion proteins.

L'invention porte sur des protéines de fusion à des anticorps avec un site de liaison à FCRN modifié et sur les molécules d'acide nucléique codantes correspondantes. La protéine de fusion à un anticorps comprend deux chaînes polypeptidiques, la première chaîne polypeptidique comprenant une molécule biologiquement, de préférence thérapeutiquement, active liée à au moins une partie d'une région constante d'immunoglobuline. La seconde chaîne polypeptidique comprend au moins une partie d'une région constante d'immunoglobuline. Une des chaînes polypeptidiques comprend une mutation dans la région constante, qui conduit à la réduction ou à la perte de la liaison à FcRn mais ne provoque pas de réduction notable de la demi-vie sérique de ladite protéine de fusion à un anticorps. Dans un mode de réalisation préféré, l'invention porte sur des molécules Fc-IFNß respectivement construites. L'invention porte également sur des procédés de production des protéines de fusion et sur des procédés d'utilisation des protéines de fusion pour traiter des maladies et des états pathologiques soulagés par l'administration des protéines de fusion.
