Provided are antigenic determinants recognized by anti-human death receptor DR5 monoclonal antibody AD5-10, derivatives and uses thereof. The antigenic determinants have the amino acid sequence of LITQQDLAPAARA, wherein the core polypeptide is QDLAP. The polypeptides comprising said antigenic determinants can activate the signal pathway downstream of DR5 after binding to monoclonal antibody AD5-10, then result in apoptosis. The antigenic determinants and derivatives thereof can be used for screening and preparing anti-human DR5 agonistic antibody, small molecular compound binding to DR5 and DR5 vaccine. Their nucleotide encoding sequences can be used for preparing antisense nucleotides and small molecular ribonucleotides for treating and preventing tumor and/or AIDS and the like.
L'invention porte sur des déterminants antigéniques reconnus par un anticorps monoclonal AD5-10 anti-récepteur de mort humain DR5, des dérivés et des utilisations de ceux-ci. Les déterminants antigéniques ont la séquence d'acides aminés de LITQQDLAPAARA, où le polypeptide central est QDLAP. Les polypeptides comprenant lesdits déterminants antigéniques peuvent activer la voie de signalisation en aval de DR5 après liaison à un anticorps monoclonal AD5-10, puis conduire à l'apoptose. Les déterminants antigéniques et dérivés de ceux-ci peuvent être utilisés pour cribler et préparer un anticorps agoniste anti-DR5 humain, un petit composé moléculaire se liant à DR5 et un vaccin contre DR5. Leurs séquences codantes nucléotidiques peuvent être utilisées pour préparer des nucléotides antisens et de petits ribonucléotides moléculaires pour traiter et prévenir une tumeur et/ou le SIDA et similaire.
