A method and antisense compound for inhibiting the growth of pathogenic bacterial cells are disclosed. The compound contains no more than 12 nucleotide bases and has a targeting nucleic acid sequence of no fewer than 10 bases in length that is complementary to a target sequence containing or within 10 bases, in a downstream direction, of the translational start codon of a bacterial mRNA that encodes a bacterial protein essential for bacterial replication. The compound binds to a target mRNA with a Tm of between 50 ~C to 60 ~C. The relatively short antisense compound are substantially more active than conventional antisense compounds having a targeting base sequence of 15 or more bases.
La présente invention concerne une technique et un composé antisens destiné à inhiber la croissance des cellules bactériennes pathogènes. Ce composé ne contient pas plus de 12 bases nucléotidiques et possède une séquence d'acides nucléiques de ciblage qui n'est pas inférieure à 10 bases dans la longueur et qui est complémentaire d'une séquence cible contenant 10 bases au maximum, dans une direction aval du codon de départ traductionnel d'un ARNm bactérien qui code une protéine bactérienne essentielle pour la réplication bactérienne. Ce composé se lie à un ARNm cible avec une Tm comprise entre 50 °C et 60 °C. Ce composé antisens relativement court est sensiblement plus actif que des composés antisens classiques possédant une séquence de bases de ciblage d'au moins 15 bases.
