The invention provides antisense antiviral compounds and methods of their use and production in inhibition of growth of viruses of the Flaviviridae, Picornoviridae, Caliciviridae, Togaviridae, Arteriviridae, Coronaviridae, Astroviridae and Hepeviridae families in the treatment of a viral infection. The antisense antiviral compounds are substantially uncharged morpholino oligonucleotides having a sequence of 12-40 subunits, including at least 12 subunits having a targeting sequence that is complementary to a region associated with stem-loop secondary structure within the 5'-terminal end 40 bases of the positive-sense RNA strand of the virus.
L'invention concerne des composés antiviraux antisens et leurs méthodes d'utilisation et de production, permettant d'inhiber la croissance de virus des familles Flaviviridae, Picornoviridae, Caliciviridae, Togaviridae, Arteriviridae, Coronaviridae, Astroviridae et Hepeviridae, pour le traitement d'une infection virale. Ces composés antiviraux antisens sont des oligonucléotides morpholino sensiblement neutres possédant une séquence comprenant 12-40 sous-unités, y compris au moins 12 sous-unités comportant une séquence de ciblage qui est complémentaire à une région associée à la structure secondaire tige-boucle dans les 40 bases de l'extrémité 5' terminale du brin d'ARN de sens positif du virus.
