A method and compound for treating skeletal muscle mass deficiency in a human subject are disclosed. The composition is an oligomer of morpholino subunits and phosphorus-containing intersubunit linkages joining a morpholino nitrogen of one subunit to a 51 exocyclic carbon of an adjacent subunit, contains between 10-40 nucleotide bases, has a base sequence effective to hybridize to an expression-sensitive region of processed or preprocessed human myostatin RNA transcript, identified, in its processed form, by SEQ ID NO:6, and is capable of uptake by target muscle cells in the subject. In practicing the method, the compound is administered in an amount and at a dosage schedule to produce an overall reduction in the level of serum Myostatin measured in the patient, and preferably to bring the myostatin level within the a range determined for normal, healthy individuals.
L'invention concerne une méthode et un composé permettant de traiter une déficience de la masse musculaire du squelette chez un sujet humain. Ladite composition est composée d'un oligomère de sous-unités morpholino et de liaisons intersous-unités contenant un phosphore reliant une azote morpholino de sous-unité à un carbone exocyclique 51 de sous-unité adjacente; contient entre 10-40 bases nucléotidiques; comprend une séquence efficace pour s'hybrider avec une région sensible à une expression de transcrit d'ARN de myostatine humaine traitée ou prétraitée, identifiée sous sa forme traitée par SEQ ID NO:6; et est capable de captage par des cellules musculaires cibles chez le sujet. Lors de la pratique de cette méthode, on administre le composé selon une quantité et une posologie permettant de produire une réduction globale du niveau de myostatine sérique mesuré chez le patient et, de préférence, de ramener ce niveau à une plage déterminée pour des individus normaux et sains.
