The subject invention relates to antisense oligonucleotides of the human Chk1 gene and to uses thereof. The human ChK1 gene is a major G2/M checkpoint gene that is activated in response to DNA damage. In particular, the gene transduces the inhibitory signal from DNA damage sensors to the basic cell cycle machinery. Thus, antisense oligonucleotides to the human Chk1 gene may be used, for example, to inhibit the gene thereby preventing G2 arrest induced by DNA damaging agents. Such antisense oligonucleotides should also further sensitize tumor cells thereby making them more sensitive to therapy than normal cells.
La présente invention concerne des oligonucléotides antisens du gène humain Chk1 et leurs utilisations. Le gène ChK1 humain est un gène important de point de contrôle G2/M qui est activé en réponse à un dommage à l'ADN. Plus particulièrement, le gène transduit le signal inhibiteur des capteurs de dommage à l'ADN au mécanisme du cycle cellulaire de base. Ainsi, on peut utiliser les oligonucléotides antisens du gène Chk1 humain, par exemple, pour inhiber le gène et empêcher ainsi l'inactivation de G2 par des agents de dommage à L'ADN. Par ailleurs, ces oligonucléotides antisens doivent sensibiliser les cellules tumorales pour les rendre plus réceptives à la thérapie que les cellules normales.
