An apparatus and method for sterilizing, seeding, culturing, storing, shipping, and testing three-dimensional tissue constructs is disclosed. The apparatus includes a fluid reservoir (18), a pump (30), at least one treatment chamber (10), and a means for controlling media flow characteristics around a tissue construct (20) disposed within the treatment chamber (10), and for controlling movement of the construct itself, so as to simulate a variety of physiologic conditions. One exemplary embodiment of the invention includes a means for applying an axial stress to the construct. Applying an axial stress to the construct during seeding and culturing results in a tissue-engineered construct with cells and their fibers oriented in a manner which is more likely to possess long term dimensional stability and the patency of native vessels with normal physiologic function.
L'invention concerne un appareil et un procédé de stérilisation, d'ensemencement, de mise en culture, de stockage, de transport et d'essai de constructions tissulaires tridimensionnelles. Ledit appareil comprend un réservoir (18) de fluide, une pompe (30), au moins une chambre de traitement (10) et un moyen pour moduler les caractéristiques d'écoulement des milieux autour d'une construction tissulaire (20) disposée dans la chambre de traitement (10) et pour commander le mouvement de la construction elle-même, de sorte qu'une variété de conditions physiologiques soient simulées. Dans un mode de réalisation présenté à titre d'exemple, un moyen pour appliquer une contrainte axiale sur la construction est prévu. L'application d'une contrainte axiale sur la construction pendant l'ensemencement et la mise en culture permet la production d'une construction à fibres, manipulée au niveau du tissu, à l'aide de cellules, et dont les fibres sont orientées de sorte que la construction soit susceptible de présenter une stabilité dimensionnelle à long terme et la perméabilité de vaisseaux endogènes ayant une fonction physiologique normale.
