An apparatus and method for sterilizing, seeding, culturing, storing, shipping, and testing vascular grafts is disclosed. Specifically, the present invention relates to an apparatus and method for seeding and culturing vascular grafts with human cells. The apparatus includes a fluid reservoir (10), a pump (12), an alternating pressure source (16), and at least one treatment chamber (14). By alternating pressure to a support structure (32) within the treatment chamber (14) upon which a vascular graft scaffold (26) is positioned, a varying radial stress is placed on the scaffold (26). This stress results in a tissue-engineered vascular graft with cells and their fibers oriented in a manner which is more likely to possess long term dimensional stability and the patency of native vessels with normal physiologic function.
La présente invention concerne un appareil et un procédé pour stériliser implanter, cultiver, stocker, transporter et tester les greffons vasculaires. Elle concerne en particulier un appareil et un procédé pour implanter et cultiver des greffons vasculaires avec des cellules humaines. L'appareil comprend un réservoir de fluide (10), une pompe (12), une source de pression (16) alternée et au moins une chambre de traitement (14). En alternant la pression exercée sur une structure support (32) dans la chambre de traitement (14) au dessus de laquelle est positionnée une greffe vasculaire servant de support (26), celle-ci (26) est soumise à une contrainte radiale variable. De cette contrainte résulte un greffon vasculaire engendré par génie tissulaire, dont les cellules et les fibres sont orientées de manière à mieux garantir la stabilité dimensionnelle à long terme et la perméabilité des vaisseaux natifs à fonction physiologique normale.
