Cystic fibrosis (CF) is the most common fatal autosomal recessive disease in the U.S. and is principally caused by the DF508 mutation in the CFTR gene. The principal site of morbidity and mortality for this disease is the lung. We have used genomic and proteomic methods to identify ubiquitin carboxy terminal hydrolase-1 (UCHL1) as a biomarker for cystic fibrosis. Both gene expression and cognate protein expression are massively upregulated in CF lung epithelial cells. We suggest that this gene can be useful in the assembly of a diagnostic or prognostic chip for CF, or as a target for therapeutic intervention.

La mucoviscidose (MV) est la maladie récessive autosomique mortelle la plus répandue aux Etats-Unis et est provoquée principalement par la mutation DF508 du gène CFTR. La première cause de morbidité et de mortalité chez les patients atteints de mucoviscidose survient lorsque la maladie touche les poumons. L'invention concerne l'utilisation de méthodes génomiques et protéomiques pour identifier l'ubiquitine hydrolase (UCH-L1) comme marqueur biologique pour la mucoviscidose. L'expression du gène et l'expression de la protéine analogue sont soumises à une régulation à la hausse dans les cellules épithéliales pulmonaires de sujets atteints de mucoviscidose. Ce gène peut être utilisé pour l'élaboration d'une puce de diagnostic ou de pronostic pour la mucoviscidose, ou comme cible pour une intervention thérapeutique.
