According to the present invention there is provided a method of producing a protein in a eukaryotic cell line, comprising the steps of a) providing a backbone of an artificial chromosome, b) recombining the nucleic acid encoding said protein into said backbone to generate an expression vector, c) introducing said expression vector into a eukaryotic host cell line to obtain a eukaryotic expression cell line, d) cultivating said expression cell line to produce said protein, and e) isolating said protein. The invention further relates to respective vectors and transgenic cell lines.

La présente invention concerne un procédé de production dune protéine dans une lignée cellulaire eucaryote, comprenant les étapes consistant à a) produire un squelette dun chromosome artificiel, b) recombiner lacide nucléique codant pour ladite protéine dans ledit squelette pour générer un vecteur dexpression, c) introduire ledit vecteur dexpression dans une lignée cellulaire hôte eucaryote pour obtenir une lignée cellulaire dexpression eucaryote, d) cultiver ladite lignée cellulaire dexpression pour produire ladite protéine, et e) isoler ladite protéine. Linvention concerne en outre les vecteurs et les lignées cellulaires transgéniques respectifs.
