The invention provides an in vitro avascular human corneal equivalent that comprises immortalized human cell lines. As these corneal equivalents are in vitro models, they are maintained in an incubator throughout the testing period, thereby eliminating the problems and expense associated with animal care. The corneal equivalent is preferably surrounded by a matrix in which angiogenesis (formation of capillary-like structures) can occur in vitro. This surrounding matrix has the potentioal to play the role of a pseudo-sclera, allowing the in vitro assessment of the cornea's angiogenic reaction to any substance or injury. Furthermore, the model is capable of being produced easily, is physiologically functional and can give predictable and quantifiable results when submitted to various drugs, chemicals and/or physical trauma. Modifications can be made, such as the use of primary donor cells instead of cell lines; and the physical and chemical treatments of the matrix material to make the constructs suitable for use in transplantation.
La présente invention concerne un équivalent in vitro de cornée humaine avasculaire comprenant des lignées cellulaires humaines immortalisées. Puisque ces équivalents cornéens humains sont des modèles in vitro, ils sont maintenus dans un incubateur pendant la période de test, ce qui permet d'éliminer les problèmes et les frais relatifs aux soins des animaux. L'équivalent cornéen est de préférence entouré d'une matrice dans laquelle peut s'opérer l'angiogenèse (formation de structures de type capillaire). Cette matrice périphérique peut jouer le rôle d'une pseudo-sclérotique, permettant ainsi l'évaluation in vitro de la réaction angiogénique de la cornée à toute substance ou blessure. De plus, on peut facilement produire ce modèle qui est par ailleurs physiologiquement fonctionnel et peut donner des résultats prévisibles et quantifiables lorsqu'il est soumis à une variété de médicaments, à un trauma chimique et/ou physiologique. On peut également apporter des modifications telles que l'utilisation de cellules primaires de donneur à la place de lignées cellulaires, et soumettre la matière matricielle à des traitements physiques et chimiques afin d'adapter les produits de synthèse à la transplantation.
