The invention relates to an artificial dermis (1) obtained from plasma containing platelets (2) and human fibroblasts (3). Said plasma with platelets (2) is obtained by fractionating the whole blood (4) of the patient (8) by means of light centrifugation and the human fibroblasts (3) are obtained from a skin biopsy (5). The gelation is obtained by adding calcium. The inventive artificial dermis (1) favours a rapid growth of the keratinocytes (6) seeded on the surface thereof in order to form an artificial skin (7) that can be easily transplanted. Large areas of artificial dermis (1) are obtained from a small skin biopsy (5) and from minimum quantities of plasma with platelets (2). Said plasma is rich in cytokines and in platelet growth factors and therefore stimulates the proliferation of the seeded cells, both internally and on the surface. The artificial skin (7) thus produced can be used for the treatment of serious burns, chronic cutaneous ulcers, etc. or as a base for gene therapy through the use of genetically modified cells.

L'invention concerne un derme artificiel (1) obtenu à partir de plasma contenant des plaquettes (2) et des fibroblastes (3) humains. On obtient ce plasma contenant des plaquettes (2) par fractionnement du sang total (4) du patient (8) au moyen d'une centrifugation légère et les fibroblastes (3) humains à partir d'une biopsie de peau (5). On obtient la gélification par addition de calcium. Ce derme artificiel (1) favorise une croissance rapide des kératinocytes (6) semés sur leur surface pour constituer une peau artificielle (7) qui est facilement transplantable. On obtient de grandes surfaces de derme artificiel (1) à partir d'une petite biopsie de peau (5) et de quantités minimes de plasma contenant des plaquettes (2). Etant donné que ce dernier est enrichi en cytokines et en facteurs de croissance plaquettaire, il stimule la prolifération des cellules semées, tant intérieurement que superficiellement. La peau artificielle (7) ainsi obtenue peut servir au traitement des grands brûlés, des ulcères cutanées chroniques ou servir de base pour une thérapie génique, à l'aide de cellules génétiquement modifiées.
