An assay system is provided for detecting the enzymatic activity of a phosphorylation enzyme, which enzyme may be a phosphatase or protein kinase. The substrate for the enzyme is immobilized on a solid support <i>via</i> covalent or non-covalent binding, through a signal enhancing polymer. The immobilized substrate provides an enhanced signal to background ratio, when compared to a substrate in solution. The methods are easily adapted to high throughput screening systems.

L'invention concerne un système de dosage destiné à la détection de l'activité enzymatique d'une enzyme de phosphorylation, cette dernière pouvant être une phosphatase ou une protéine kinase. Le substrat de l'enzyme est immobilisé sur un support solide via une liaison covalente ou non-covalente, par l'intermédiaire d'un polymère d'amplification de signal. Le substrat immobilisé produit un signal amplifié par rapport au rapport de fond, lorsqu'il est comparé à un substrat de la solution. Ces méthodes sont facilement adaptées à des systèmes de criblage à débit élevé.
