A non-immunochemical method is disclosed for determining homocysteine in a sample. A combination of the sample and a first compound that comprises a cis- 1,4-dioxo-2-butene moiety or hydrolytically derived precursor thereto is provided in a liquid medium. The combination is subjected to conditions under which homocysteine reacts with the cis-1,4-dioxo-2-butene moiety or hydrolytically derived precursor thereto to form a product, which is then detected without separation of the product from the medium. A second compound such as a mercaptan containing reagent or an antibody for cysteine can optionally be included to facilitate discrimination of the product formed from homocysteine from products formed from other amines in the sample. The product from the reaction of homocysteine has a six-member ring fused to an unsaturated five-member ring. The junction of the rings is comprised of the nitrogen of homocysteine and an atom that comprises a double bond. The six- member ring comprises the sulfur atom of homocysteine. The amount of the product detected is related to the amount of homocysteine in the sample.
L'invention concerne une méthode non-immunochimique de détermination d'une homocystéine dans un échantillon. On réalise une combinaison, dans un milieu liquide, de l'échantillon et d'un premier composé comprenant une fraction de cis-1,4-dioxo-2-butène ou son précurseur dérivé par hydrolyse. La combinaison est soumise aux conditions dans lesquelles une homocystéine entre en réaction avec la fraction de cis-1,4-dioxo-2-butène ou son précurseur dérivé par hydrolyse pour former un produit que l'on détecte ensuite sans séparer le produit du milieu. On peut éventuellement introduire un deuxième composé tel qu'un réactif contenant du mercaptan ou un anticorps contre la cystéine pour faciliter la distinction entre le produit provenant de l'homocystéine et des produits provenant d'autres amines dans l'échantillon. Le produit issu de la réaction d'homocystéine présente un noyau à six éléments réuni par fusion avec un noyau à cinq éléments non saturé. La jonction des noyaux est formée de l'azote de l'homocystéine et d'un atome comportant une double liaison. Le noyau à six éléments comprend l'atome de soufre de l'homocystéine. La quantité de produit détectée est relative à la quantité d'homocystéine dans l'échantillon.
