The invention provides methods for detecting the presence of a non-prion pathogenic conformer in a sample by contacting the sample suspected of containing a non-prion pathogenic conformer with a pathogenic conformer- specific binding reagent under conditions that allow the binding of the reagent to the pathogenic conformer, if present; and detecting the presence the pathogenic conformer, if any, in the sample by its binding to the reagent; where the pathogenic conformer specific binding reagent is typically derived from a prion protein fragment and interacts preferentially with a pathogenic prion protein. Methods for diagnosis of conformational diseases are also provided.

L'invention concerne des procédés pour détecter la présence d'un conformère pathogène non prion dans un échantillon par mise en contact de l'échantillon susceptible de contenir un conformère pathogène non prion avec un réactif de liaison spécifique pour un conformère pathogène dans des conditions qui permettent la liaison du réactif avec le conformère pathogène, s'il est présent; et détection de la présence éventuelle du conformère pathogène dans l'échantillon par sa liaison avec le réactif. Le réactif de liaison spécifique pour un conformère pathogène est de manière typique dérivé d'un fragment de protéine prion et interagit de manière préférentielle avec une protéine prion pathogène. Des procédés de diagnostic de maladies conformationnelles sont également décrits.
