In an assay, an analyte is specifically associated with a reporter adenylate kinase, ADP is added and then formation of ATP is mo nitored. Prior to addition of ADP, adenylate kinase other than reporter adenylate kinase is removed. Assay apparatus comprises a solid phase on which is immobilised the analyte or an antibody specific for the analyte, a reporter composition comprising a thermostable adenylate kinase coupled to an antibody specific for the analyte, and ADP plus associated reagents for conversion of ADP into ATP by thermostable adenylate kinase.
L'invention concerne un dosage dans lequel on associe de façon spécifique un analyte à une adénylate kinase rapporteur, on ajoute de l'ADP et on surveille ensuite la formation d'ATP. Avant d'ajouter l'ADP, on retire une adénylate kinase autre que l'adénylate kinase rapporteur. L'appareil servant au dosage comporte une phase solide sur laquelle on immobilise l'analyte ou un anticorps spécifique à l'analyte, une composition rapporteur contenant une kinase adénylate thermostable couplée à un anticorps spécifique à l'analyte, et de l'ADP et des réactifs associés pour la conversion de l'ADP en ATP au moyen d'une kinase adénylate thermostable.
