The present invention provides cell-based assays for inositol phosphates involving the preferential binding of radiolabeled inositol phosphates to a solid phase containing a scintillant within. The assay allows one to screen for inhibitors of inositol phosphate phosphatases or GPCRs which are coupled to phosphoinositide hydrolysis. The assays are fast, convenient, and avoid the column chromatography steps that prior art methods employed.
La présente invention concerne des dosages à base de cellules destinés à des phosphates d'inositol impliquant la liaison préférentielle de phosphates d'inositol identifiés au moyen de marqueur radioactif, à une phase solide renfermant un agent scintillant. Ledit dosage permet de cribler des inhibiteurs d'inositol phosphate phosphatases ou des GPCR qui sont couplés à l'hydrolyse des phosphoïnositides. Les dosages sont rapides, adéquats et évitent les étapes de chromatographie en colonne que les méthodes de la technique antérieure utilisaient.
