A C to T DNA variation at position 3365 in exon 5 of the human Asthma Associated Factor 1(AAF1) produces the predicted amino acid substitution of a methionine for a threonine at codon 117 of AAF1. When this substitution occurs in both alleles in one individual, it is associated with less evidence of atopic allergy including asthma, fewer abnormal skin test responses, and a lower serum total IgE. Thus, applicant has identified the existence of a non-asthmatic, non-atopic phenotype characterized by methionine at codon 117 when it occurs in both AAF1 gene products in one individual.
Dans l'exon 5 du facteur 1 associé à l'asthme humain, une transition de T à C de l'acide nucléique de la position 3365 produit la substitution prévue des acides aminés, celle d'une méthionine qui remplace une thréonine, au codon 117 de ce facteur. Quand cette substitution intervient dans les deux allèles d'un même sujet, elel est associée à une prévalence moindre des allergies atopiques, y compris l'asthme, à moins de réactions anormales aux tests cutanés, et à un niveau total moindre de l'IgE sérique. Le déposant a donc identifié l'existence d'un phénotype, non lié à l'asthme et non atopique, caractérisé par la méthionine du codon 117 lorsqu'elle est présente dans les deux produits géniques du facteur 1 associé à l'asthme humain chez un même sujet.
