Reverse transcription of retroviruses is initiated from a 18 nucleotide primer binding site (PBS), located within the 5' region of viral genomic RNA, to which the host cell-derived tRNA primer is annealed and also involves viral genomic sequences outside the PBS. Significantly lower levels of viral DNA were detected in cells infected with proviral DNA clones of human immunodeficiency virus (HIV) selectively deleted in regard to a segment found immediately downstream of the PBS. This segment is involved in efficient expression of each of viral DNA, mRNA, and infectious virus. The deleted DNA clones are useful for the preparation of an HIV vaccine.
La transcription inverse des rétrovirus est initiée à partir d'un site de fixation d'une amorce (PBS) de 18 nucléotides, situé à l'extrémité 5' de l'ARN génomique viral et auquel l'amorce d'ARNt dérivé de la cellule-hôte est annelée, et met également en jeu des séquences génomiques virales à l'extérieur du PBS. On a constaté une concentration significativement inférieure d'ADN viral dans les cellules infectées par les clones ADN proviral du virus de l'immunodéficience humaine (VIH) amputés sélectivement d'un segment situé immédiatement en aval du PBS. Ce segment joue un rôle dans l'expression efficace de l'ADN viral, de l'ARNm viral et du virus infectieux. Les clones ADN amputés sont utiles dans la préparation d'un vaccin contre le VIH.
