This application relates to the use of attenuated prokaryotic microorganism strains (such as Salmonella) expressing nucleic acid encoding HPV proteins as vaccines against HPV infection and the associated increased risk of cancer. In particular, the work shows that it is possible to assemble VLPs in a prokaryotic organism and that nasal immunization of mice with the strains HPV- specific conformationally dependent and neutralizing antibodies in serum and genital secretions. The experiments described herein show that it is also possible to assemble chimeric VLPs of an HPV including a fusion partner and that tumor protection can be induced.

Cette invention concerne l'utilisation de souches atténuées de micro-organismes procaryotiques (tels que Salmonella), exprimant un acide nucléique codant des protéines du VPH, comme vaccins contre l'infection à VPH et le risque accru de cancer accompagnant ces infections. Ce travail montre en particulier qu'il est possible d'assembler des particules de type viral dans un organisme procaryotique et que l'immunisation par voie nasale de souris à l'aide de ces souches fait apparaître des anticorps spécifiques du VPH, de conformation dépendante et neutralisants dans le sérum et les sécrétions génitales. Les expériences décrites montrent qu'il est également possible d'assembler des particules de type viral chimères d'un VPH en incluant un partenaire de fusion et qu'il est possible d'induire une protection anti-tumorale.
