The invention relates to novel live strains which are intended for oral immunisation against cholera and which are lyophilised in formulations enabling the long-term storage thereof. The aforementioned strains combine the two most important properties that known vaccine candidates must have for the purpose of oral immunisation against cholera, namely: a level of reactogenicity which is reduced to acceptable levels through the use of known mutations which have been described in prior art; and an improved environmental safety index owing to the suppression of the mshA gene or spontaneous mutations which result in the absence of the MSHA pilus on the outer surface of the bacteria and the absence of the VGJF phage in the gene pool thereof, which, if present, enable the incorporation and dissemination of the CTXF phage assisted by VGJF.
La présente invention concerne de nouvelles souches vivantes utilisées par voie orale pour immuniser un individu contre le choléra, lyophilisées en préparations permettant un stockage à long terme. Ces souches contiennent les deux propriétés les plus importantes que doivent posséder les candidats vaccinaux destinés à l'immunisation contre le choléra. Une de ces propriétés est définie par un niveau de réactogénicité réduit jusqu'à des niveaux acceptables, en raison de l'utilisation de mutations déjà connues et décrites dans les antériorités. L'autre propriété est définie par un meilleur indice de protection environnementale en raison de la suppression du gène mshA ou de mutations spontanées conduisant à l'absence du pilus MSHA sur la surface externe de la bactérie et à l'absence du phage VGJ.PHI. dans sa dotation génétique qui, s'ils étaient présents, permettraient l'introduction et la dissémination du phage CTX.PHI. assistée par VGJ.PHI..
