A method for regulating in vivo calcium transport in cardiac muscle of animals suffering from congestive heart failure is disclosed. According to the method, calcium ATPase activity (which decreases as congestive heart failure develops) and cardiac muscle contractility augmented by delivering a gene which operatively encodes the enzyme into the heart. Delivery systems (including recombinant expression vectors) are provided, as are methods for monitoring the expression and effect of the gene product on cardiac performance.

Ce procédé permet de moduler in vivo le transport du calcium dans le muscle cardiaque d'animaux souffrant de défaillance cardiaque congestive. Il consiste à accroître l'activité de la calcium ATPase (qui diminue quand une défaillance cardiaque congestive survient) et la contractilité du muscle cardiaque par l'apport d'un gène qui code effectivement cette enzyme à destination du coeur. On décrit des vecteurs de libération (y compris des vecteurs d'expression recombinés), ainsi que des procédés permettant de contrôler l'expression d'un tel produit génique et son effet sur le rendement du coeur.
