A system and method for rapidly detecting cells (1) of interest using multi- dimensional, highly quantitative, nuclear and cytoplasmic densitometric features (NDFs and CDFs) includes a flow optical tomography (FOT) instrument capable of generating various optical projection images (or shadowgrams) containing accurate density information from a cell, a computer (40) and software to analyze and reconstruct the projection images into a multi- dimensional data set, and automated feature collection and object classifiers. The system and method are particularly useful in the early detection of cancers such as lung cancer using a bronchial specimen from sputum or cheek scrapings and cervical/ovarian cancer using a cervical scraping, and the system can be used to detect rare cells in specimens including blood.

L'invention concerne un système et un procédé de détection rapide de cellules d'intérêt au moyen de caractéristiques densitométriques nucléaires et cytoplasmiques multidimensionnelles, fortement quantitatives. Ledit système est composé d'un instrument de tomographie optique à flux pouvant produire plusieurs images de projection optique (ou hologrammes à transformée d'ombre) contenant des informations de densité précises d'une cellule, d'un ordinateur et d'un logiciel destiné à analyser et reconstruire les images de projection en un ensemble de données multidimensionnel, et d'éléments de récupération de caractéristiques et de classification d'objets. Le système et le procédé selon l'invention servent notamment à un dépistage précoce de cancers tels que le cancer des poumons à partir d'échantillons bronchiques d'expectorations ou de prélèvements jugaux, et de dépistage de cancers tels que le cancer du col de l'utérus ou de l'ovaire à partir de prélèvements cervicaux, et à la détection de cellules rares dans des échantillons tels que des échantillons de sang.
