ABSTRACT A temperature sensitive monomeric bacterial luciferase enzyme is created through the use of site specific mutagenesis and recombinant DNA technology such that the expression of the DNA encoding for the enzyme is placed under the control of a single promoter and the two wild type genes all converted to a single reading frame. A method for detecting the presence or absence of protein folding regulatory and control proteins in a cell transformed with a vector containing the DNA encoding for the temperature sensitive luciferase enzyme is also provided.
