A method and apparatus for representing double stranded nucleic acid fragments which have been separated by a chromatographic process as an array of bands which can be accurately quantified, optimized and stored. Using, for example, a Matched Ion Polynucleotide Chromatography (MIPC) process, an analog output from a UV detector is digitized and input to a computer. The digitized signal is converted to a linear array of bands which may be displayed on a video display terminal. The intensity and/or color of a band may correlate to the amount of double stranded nucleic acid in the respective fraction or the respective double stranded nucleic acid fragment above a user selected threshold level at a corresponding point in the digitized signal. The calculated base pair length, concentration, and retention time of each band in the array of bands may be displayed in alphanumeric form.
L'invention a pour objet un procédé et un appareil pour représenter des fragments d'acides nucléiques à double brin, séparés par un procédé de chromatographie, sous la forme d'un arrangement de bandes qui peuvent être mesurées, optimisées et stockées de manière précise. En utilisant, par exemple, un procédé de chromatographie des polynucléotides par ions appariés (MIPC), on numérise une sortie analogique provenant d'un détecteur ultraviolet et on l'introduit dans un ordinateur. Le signal numérisé est converti en un arrangement linéaire de bandes qui peut être affiché sur un terminal vidéo. L'intensité et/ou la couleur d'une bande peut être en corrélation avec la quantité d'acide nucléique à double brin dans la fraction respective ou le fragment respectif de l'acide nucléique à double brin dépassant la valeur du seuil, sélectionnée par l'utilisateur, au niveau d'un point correspondant du signal numérisé. Il est possible d'afficher sous forme alphanumérique la longueur calculée d'une paire de bases, la concentration et le temps de rétention dans chaque bande faisant partie de l'arrangement de bandes.
