Using data electrophoretic trace data from conventional nucleic acid sequencing equipment, a method for basecalling that is tolerant to variable peak spacing is described. The method generates high-quality basecalls and reliable quality scores. In addition, a new type of quality score that estimates the probability of a deletion error between the current and the following basecall is described. A new protocol for benchmarking that better discerns basecaller performance is also provided.

L'invention concerne l'utilisation de données de traces électrophorétiques tirées d'un matériel classique de séquençage d'acide nucléique dans un procédé de conversion de données brutes en une séquence de bases tolérant l'espacement de crête variable. Le procédé génère des séquences de base de haute qualité et permet d'obtenir des scores de qualité fiable. De plus, l'invention concerne un nouveau type de score de qualité estimant la probabilité d'une erreur de délétion entre la séquence de bases courante et la séquence de bases suivante. L'invention concerne également un nouveau protocole d'étalonnage discernant mieux l'efficacité d'unités de conversion de données en séquences de bases.
