A method is provided of making meltable beads for use in reactions for the amplification and/or synthesis of a polynucleotide, the method comprising (i) Providing one or more reagents for use in a reaction for the amplification and/or synthe-sis of a polynucleotide and a substance for promoting the forma-tion of a solid bead; (ii) Providing a reactor device comprising a sample conduit and a first carrier fluid conduit for the flow of immiscible liquids, the sample conduit and first carrier fluid con-duit meeting at a junction; (iii) Heating the one or more reagents for use in a reaction for the amplification and/or synthesis of a polynucleotide and the substance for promoting the formation of a solid bead so as to form a liquid comprising the one or more reagents for use in a reaction for the amplification and/or syn-thesis of a polynucleotide and the substance for promoting the formation of a solid bead in intimate admixture; (iv) Passing the liquid down the sample conduit and a carrier fluid down the first carrier fluid conduit, thus causing the formation of droplets at or downstream of the junction and (v) Causing the formation of solid beads by cooling the droplets. A device for use in such methods is also provided, as is a plurality of beads made by such methods.
L'invention concerne un procédé de fabrication de billes fusibles à utiliser dans des réactions d'amplification et/ou de synthèse d'un polynucléotide. Ledit procédé comprend les opérations consistant à (i) se procurer un ou plusieurs réactifs destinés à être utilisés dans une réaction d'amplification et/ou de synthèse d'un polynucléotide et une substance pour favoriser la formation d'une bille solide ; (ii) se procurer un dispositif de réacteur comprenant un conduit d'échantillon et un premier conduit de fluide support pour l'écoulement de liquides non miscibles, le conduit d'échantillon et le premier conduit de fluide support se rencontrant à une jonction ; (iii) chauffer le ou les réactifs pour utilisation dans une réaction d'amplification et/ou de synthèse d'un polynucléotide et la substance pour favoriser la formation d'une bille solide de façon à former un liquide comprenant le ou les réactifs destinés à être utilisés dans une réaction d'amplification et/ou de synthèse d'un polynucléotide et la substance pour favoriser la formation d'une bille solide en mélange intime ; (iv) faire descendre le liquide dans le conduit d'échantillon et un fluide support dans le premier conduit de fluide support, provoquant ainsi la formation de gouttelettes en aval ou à la jonction et (v) provoquer la formation de billes solides par refroidissement des gouttelettes. L'invention concerne également un dispositif destiné à être utilisé dans de tels procédés, de même qu'une pluralité de billes obtenues par de tels procédés.
