The invention concerns benzimidazole derivatives of general formula (I) wherein: X represents a nitrogen atom or a carbon atom, and when X represents a nitrogen atom: R3 represents a hydrogen atom or a C¿1?-C¿4? alkyl group, or does not exist to produce compounds of formula (I) comprising a secondary or tertiary amine; R4 represents a hydrogen atom or a C¿1?-C¿6? alkyl, C¿3?-C¿7? cycloalkyl, C¿3?-C¿7? heterocycloalkyl group optionally substituted, -(CH¿2?)¿p?-heteroaryl, heteroarylcarbonyl, phenylcarbonyl, C¿1?-C¿6? alkylcarbonyl, -(CH¿2?)¿p?COOR, phenylsulphonyl group optionally substituted; and when X represents a carbon atom: R3 represents a hydrogen atom or a -NR5R6, -N(R5)¿3??+¿, -NHCOR7, -CONHR5, -COR7, -NHCONH¿2?, -OH or CH¿2?OH group, R4 represents a hydrogen atom or a -(CH¿2?)¿p?-phenyl group optionally substituted, -(CH¿2?)¿p?-heteroaryl or-(CH¿2?)¿t?NR7R8 group. The invention is useful in therapeutics for preparing a medicine for preventing or treating disorders wherein the poly(ADP-ribose) polymerase is involved.
L'invention concerne des dérivés de benzimidazole de formule générale (I) dans laquelle X représente un atome d'azote ou un atome de carbone; et dans le cas oú X représente un atome d'azote: R3 représente un atome d'hydrogène ou un groupe (C1-C4)alkyle, ou n'existe pas pour donner des composés de formule (I) comportant une amine secondaire ou tertiaire; R4 représente un atome d'hydrogène ou un groupe (C1-C6)alkyle, (C3-C7)cycloalkyle, (C3- C7)hétérocycloalkyle éventuellement substitué, -(CH2)p-hétéroaryle, hétéroarylcarbonyle, phénylcarbonyle, (C1-C6)alkylcarbonyle, -(CH2)pCOOR, phénylsulfonyle éventuellement substitué ou -(CH2)P-phényle éventuellement substitué, et dans le cas oú X représente un atome de carbone: R3 représente un atome d'hydrogène ou un groupe -NR5R6, -N(R5)3+, -NHCOR7, -CONHR5, COR7, - NHCONH2, -OH ou -CH2OH, R4 représente un atome d'hydrogène ou un groupe - (CH2)p-phényle éventuellement substitué, -(CH2p-hétéroaryle ou -(CH2)tNR7R8. Application en thérapeutique pour la préparation d'un médicament destiné à la prévention ou au traitement des désordres dans lesquels l'enzyme poly(ADP- ribose) polymérase est impliquée.
