The present invention is concerned with vasoconstrictive [(benzodioxan, benzofuran or benzopyran)alkylamino]alkyl substituted guanidines having formula (I), the pharmaceutically acceptable acid addition salts thereof, and the stereochemically isomeric forms thereof, wherein X is O, CH2 or a direct bond; R1 is hydrogen or C1-6alkyl; R2 is hydrogen, C1-6alkyl, C3-6alkenyl or C3-6alkynyl; R3 is hydrogen or C1-6alkyl; or R2 and R3 may be taken together to form a bivalent radical of formula -(CH2)m-, wherein m is 4 or 5; or R1 and R2 taken together may form a bivalent radical of formula -CH = CH- or of formula -(CH2)n-, wherein n is 2, 3 or 4; or R3 may represent a bond when R1 and R2 taken together form a bivalent radical of formula -CH=CH-CH=, -CH=CH-N=, or -CH=N-CH=; R4 is hydrogen or C1-6alkyl; Alk1 is a bivalent C1-3 alkanediyl radical; A is a bivalent radical of formula (a), (b), (c), (d), (e), wherein each R5 is hydrogen or C1-4alkyl; wherein each R6 is hydrogen or C1-4alkyl; Alk2 is C2-15alkanediyl or C5-7cycloalkanediyl; and each p is 0, 1 or 2; provided that [2-[2,3-dihydro- 1,4-benzodioxin-2-yl)methyl]amino]ethyl guanidine is excluded. Pharmaceuticals which are useful as vasoconstrictors. Compositions comprising said guanidine derivatives as active ingredients, processes for preparing said guanidine derivatives and novel N-cyano guanidine, intermediates; and a use as a medicine are described.
Guanidines vasoconstrictrices substituées par [(benzodioxanne, benzofuranne ou benzopyranne)alkylamino]alkyle de la formule (I), leurs sels d'addition d'acide pharmaceutiquement acceptables, et leurs formes stéréochimiquement isomères. Dans la formule (I), X représente O, CH2 ou une liaison directe; R1 représente hydrogène ou alkyle C1-6; R2 représente hydrogène, alkyle C1-6, alcényle C3-6 ou alcynyle C3-6; R3 représente hydrogène ou alkyle C1-6; ou R2 et R3 peuvent être pris ensemble pour former un radical bivalent de la formule -(CH2)m-, dans laquelle m représente 4 ou 5; ou R1 et R2, pris ensemble, peuvent former un radical bivalent de la formule -CH=CH- ou de la formule -(CH2)n-, dans laquelle n représente 2, 3 ou 4; ou R3 peut représenter une liaison lorsque R1 et R2, pris ensemble, forment un radical bivalent de la formule -CH=CH-CH=, -CH=CH-N= ou -CH=N-CH=; R4 représente hydrogène ou alkyle C1-6; Alk1 représente un radical alcaneC1-3diyle bivalent; A représente un radical bivalent des formules (a), (b), (c), (d) ou (e), dans lesquelles chaque R5 représente hydrogène ou alkyle C1-4; chaque R6 représente hydrogène ou alkyle C1-4; Alk2 représente alcaneC2-15diyle ou cycloalcaneC5-7diyle; et chaque p représente 0, 1 ou 2; à l'exception de [2-[(2,3-dihydro-1,4-benzodioxin-2-yl)méthyl]amino]éthyl guanidine; produits pharmaceutiques utilisables comme vasoconstricteurs; compositions comportant lesdits dérivés de guanidine à titre d'ingrédients actifs; procédés de préparation desdits dérivés de guanidine; nouveaux intermédiaires de N-cyano guanidine; et applications médicamenteuses.
