Disclosed are benzoxazines and benzothiazines of formula (I) wherein Q is O- (CHR6)1-3 or S-(CHR6)1-3, R1 and each R 6 is H, optionally substituted [C1-6 alkyl, C1-4alkaryl, C1-4 alkheterocyclyl, C2-9 heterocyclyl, C3-8 cycloalkyl, C1-4 alkcy- cloalkyl] or -(CR1A R1B)nNR1c R1D wherein R1A R1B R1C R1D are independently hydrogen, optionally substituted [C1-6 alkyl, C1-4alkaryl, Cl-4 alkheterocyclyl, C2-9 heterocyclyl, C3-8 cycloalkyl, etc ] or wherein R1A and R1B combine to form =0, or wherein R1C and R1D combine to form an optionally substituted C2-9 heterocyclyl, and n is an integer between 1-6, each of R2 and R3 is H, hal, optionally substituted [C1-6 alkyl, C6-10aryl, C1-6alkaiyl, C2-9 heterocyclyl, C1-6 alkoxy, C1-6 thioalkoxy, (CH2)r2NHC(NH)R2A, CH2)r2NHC(S)NHR2A, Cl-4 alkheterocyclyl] or hydroxy, wherein r2 is an integer from 0 to 2, R2A is optionally substituted [C1-6 alkyl, C6-10aryl, C^alkaryl, C2-9 heterocyclyl, C1-4 alkheterocyclyl,C1-6 thioalkoxy, Cl-4 thioalkaryl, aryloyl, Cl-4thioalkheterocylyl, or amino], each R4 and R5 is H, hal, (CH2)r2NHC(NH)R2A or CH2)r2NHC(S)NH R2A, Y' and Y 2 are together =0 or Y1 and Y2 are independently H, optionally substituted [C1-6 alkyl, C6-10aryl, C1-6alkaryl, C2-9 heterocyclyl, C1-6 alkoxy, C1-6 thioalkoxy, C1-4alkheterocyclyl] or hydroxy; wherein only one of R2 , R3, R4 and R5 is (CH2)r2NHC(NH)R2A or CH2)r2NHC(S)NH R2A, or a pharmaceutically acceptable salt or prodrug thereof Said benzoxazines and benzothiazines of formula (I) inhibit nitric oxide synthase (N O S), particularly selectively inhibit neuronal nitric oxide synthase (n N O S) in preference to other N O S iso-forms The N O S inhibitors of formula (I), alone or in combinatioon with other pharmaceutically active agents, can be used for treating or preventing various medical conditions
L'invention porte sur des benzoxazines et des benzothiazines de formule (I) dans laquelle Q représente O-(CHR6)1-3 ou S-(CHR6)1-3 ; R1 et chaque R6 représentent H, [alkyle en C1-6, (alkyl en C1-4)aryle, (alkyl en C1-4)hétérocyclyle, hétérocyclyle en C2-9, cycloalkyle en C3-8, (alkyl en C1-4)cycloalkyle] éventuellement substitués ou -(CR1AR1B)nNR1cR1D où R1A, R1B, R1C, R1D représentent indépendamment hydrogène, [alkyl en C1-6, (alkyl en C1-4)aryle, (alkyl en C1-4)hétérocyclyle, hétérocyclyle en C2-9, cycloalkyle en C3-8, etc.] éventuellement substitués ou où R1A et R1B se combinent pour former =O, ou où R1C et R1D se combinent pour former un hétérocyclyle en C2-9 éventuellement substitué, et n représente un entier entre 1 et 6 ; chacun parmi R2 et R3 représente H, hal, [alkyle en C1-6, aryle en C6-10, (alkyl en C1-6)aryle, hétérocyclyle en C2-9, alcoxy en C1-6, thioalcoxy en C1-6, (CH2)r2NHC(NH)R2A, (CH2)r2NHC(S)NHR2A, (alkyl en C1-4)hétérocyclyle] éventuellement substitués ou hydroxy, où r2 représente un entier de 0 à 2, R2A représente [alkyl en C1-6, aryle en C6-10, (alkyl en C1-6)aryle, hétérocyclyle en C2-9, (alkyl en C1-4)hétérocyclyle, thioalcoxy en C1-6, (thioalkyl en C1-4)aryle, aryloyle, (thioalkyl en C1-4)hétérocyclyle, ou amino] éventuellement substitués ; chaque R4 et R5 représente H, hal, (CH2)r2NHC(NH)R2A ou (CH2)r2NHC(S)NH R2A ; Y1 et Y2 représentent ensemble =O ou Y1 et Y2 représentent indépendamment H, [alkyle en C1-6, aryle en C6-10, (alkyl en C1-6)aryle, hétérocyclyle en C2-9, alcoxy en C1-6, thioalcoxy en C1-6, (alkyl en C1-4)hétérocyclyle] éventuellement substitués ou hydroxy ; où seul l'un parmi R2, R3, R4 et R5 représente (CH2)r2NHC(NH)R2A ou (CH2)r2NHC(S)NH R2A ; ou un sel pharmaceutiquement acceptable ou promédicament de celles-ci. Lesdites benzoxazines et benzothiazines de formule (I) inhibent l'oxyde nitrique synthase (NOS), en particulier elles inhibent sélectivement l'oxyde nitrique synthase neuronale (NOSn) de préférence à d'autres isoformes de NOS. Les inhibiteurs de NOS de formule (I), seuls ou en association avec d'autres agents pharmaceutiquement actifs, peuvent être utilisés pour le traitement ou la prévention de diverses affections médicales.
