Provided herein are carbohydrate complement-modified bifunctional glycoproteins, and their use in tumor--selective therapy. The bifunctional glycoproteins comprise a first component that specifically binds to a tumor-specific antigen and a second component having enzymic activity by means of which a non-toxic prodrug is cleaved into a cytotoxic drug. The carbohydrate complement comprises at least one exposed carbohydrate residue selected from the group consisting of mannose, galactose, N-acetylglucosamine, lactose and fucose. The modified carbohydrate complement contributes to increased relative concentration of the glycoproteins at the site of the tumor, and enhanced clearance from the general circulation and non-tumor sites.
La présente invention porte sur des glycoprotéines bifonctionnelles modifiées munies d'un complément glucidique et leur utilisation pour traiter de manière sélective les tumeurs. Les glycoprotéines bifonctionnelles sont constituées d'une première composante que se lie spécifiquement à un antigène propre à la tumeur et à une deuxième composante présentant une activité enzymatique grâce à laquelle un promédicament non toxique est clivé pour donner un médicament cytotoxique. Le complément glucidique compred au moins un résidu glucidique exposé choisi parmi les suivants : mannose, galactose, N-acétylglucosamine, lactose et fucose. Le complément glucidique modifié contribue à hausser la concentration relative des glycoprotéines à l'endroit de la tumeur et à favoriser l'élimination de la circulation générale et des sites sans tumeur.
