A method for conjugation of glycosylated Fc-containing proteins is described. The method comprises carbamate chemistry performed and neutral pH or below and the resulting Fc-containing protein are expected to retain tertiary structure and therefore Fc-related bioactivity such as FcR binding, the ability to bind C1q, in addition to retaining ligand binding capabilities related to the incorporation of a ligand binding peptide or other polypeptide which has binding specificity. The method is exemplified using an erythropoietin-mimetic peptide fused to a human IgG1 antibody constant domain comprising a hinge, CH2 and CH3.

La présente invention concerne un procédé pour conjuguer des protéines contenant Fc glycosylées. Le procédé fait intervenir la chimie des carbamates dans des conditions de pH neutre ou en-dessous de celui-ci, et les protéines contenant Fc ainsi obtenues sont prévues pour conserver une structure tertiaire et ainsi conserver l'activité biologique liée à Fc telle que la liaison FcR, la capacité de liaison à C1q, en plus de conserver des aptitudes de liaison aux ligands en relation avec l'incorporation d'un peptide de liaison aux ligands ou autre polypeptide qui a une spécificité de liaison. Dans un exemple de procédé donné, on emploie un peptide érythropoïétine-mimétique fusionné avec un domaine constant d'anticorps IgG1 humain comprenant une région charnière, CH2 et CH3.
