A bioconjugate comprising a fluorescer (P) linked to a quencher (Q) by a tether (T) is provided. The tether (T) includes a segment that can recognize and interact with a target biomolecule. In the absence of a specific interaction of the bioconjugate with an enzyme or other target biomolecule recognizing the bioconjugate, the fluorescence of the polymer (P) is attenuated or modified by the relatively close proximity thereto of the quencher (Q). As a consequence of the association of the bioconjugate with the target biomolecule, a reaction can occur which results in a cleavage of the bioconjugate tether and a release of the fluorescent polymer and/or quencher. This sequence of events can be followed by an enhancement or amplification of the polymer fluorescence.
L'invention concerne un bioconjugué comprenant un fluorescer (P) lié à un quencher (Q) par un liant (T). Celui-ci (T) comprend un segment pouvant reconnaître une biomolécule cible et interagir avec celle-ci. En l'absence d'une interaction spécifique du bioconjugué avec une enzyme ou une autre biomolécule cible reconnaissant le bioconjugué, la fluorescence du polymère (P) est atténuée ou modifiée par la proximité relativement rapprochée de celui-ci par rapport au quencher (Q). L'association du bioconjugué et de la biomolécule cible engendre une réaction résultant en un clivage du liant du bioconjugué et une libération du polymère fluorescent et/ou du quencher. Cette séquence d'événements peut être suivie d'une amélioration ou d'une amplification de la fluorescence polymère.
