The invention involves using a plasma to functionalize a surface of medical devices such as catheters, particularly intracardial catheters, with covalently bound thermochemically reactive groups. The surface is further contacted with a bioactive agent, particularly antithrombogenic coatings, which is thermochemically covalently coupled to the reactive group to form a therapeutically effective coating. The selected bioactive agent is then covalently bound to the surface by thermochemical reaction with the surface reactive groups. In another embodiment, the functionalizing step comprises contacting the surface with a Langmuir-Blodgett film comprising an amphipathic compound and a bioactive agent and using a plasma to covalently crosslink the Langmuir-Blodgett film to the medical device surface and to the bioactive agent. Medical devices prepared by the subject methods are also provided.
Dans cette invention, on utilise un plasma pour fonctionnaliser une surface de dispositifs médicaux, tels que des cathéters et, plus particulièrement, des cathéters intracardiaques, avec des groupes thermochimiquement réactifs liés par covalence. La surface du dispositif médical en question est aussi mise en contact avec un agent bioactif, constitué particulièrement par des revêtements antithrombogènes, cet agent étant thermochimiquement couplé par covalence au groupe réactif, afin de former un revêtement thérapeutiquement efficace. L'agent bioactif choisi est ensuite lié par covalence à la surface du dispositif médical par réaction thermochimique avec les groupes réactifs de surface. Dans une autre mode de réalisation, l'étape de fonctionnalisation consiste à mettre la surface du dispositif médical en contact avec un film de Langmuir-Blodgett comprenant un composé amphipathique et un agent bioactif, et à utiliser un plasma pour fixer par réticulation par covalence ce film de Langmuir-Blodgett sur la surface du dispositif médical et sur l'agent bioactif. Des dispositifs médicaux préparés par les procédés de cette invention sont également proposés.
