The invention relates to an implant obtained by assembling in vitro various elements in order to form a neo-organ which is introduced preferably in the peritoneal cavity of the recipient. The implant comprises a biocompatible support intended to the biological anchoring of cells; cells having the capacity of expressing and secreting naturally or after recombination a predetermined compound, for example a compound having a therapeutical interest; and a constituent capable of inducing and/or promoting the geling of said cells. The invention also relates to a kit for the preparation of the implant as well as to a new recombinant retroviral vector comprising a provirus DNA sequence modified in that the genes gag, pol and env have been deleted at least partially so as to obtain a proviral DNA capable of replication. The invention also relates to recombinant cells comprising the new retroviral vector.
L'invention concerne un implant obtenu par l'assemblage in vitro de différents éléments afin de former un néo-organe qui est introduit de préférence dans la cavité péritonéale du receveur. L'implant comprend: un support biocompatible permettant l'ancrage biologique de cellules; des cellules ayant la capacité d'exprimer et de sécréter naturellement ou après recombinaison un composé déterminé, par exemple un composé ayant un intérêt thérapeutique; et un constituent capable d'induire et/ou de favoriser la gélification desdites cellules. L'invention concerne également un kit pour la préparation de l'mplant ainsi qu'un nouveau vecteur rétroviral recombinant comprenant une séquence d'ADN de provirus modifiée en ce que les gènes gag, pol et env ont été délétés au moins en partie et ce afin d'obtenir un ADN proviral capable de se répliquer. L'invention concerne les cellules recombinantes comprenant le nouveau vecteur rétroviral.
